BulletinJuly 2020• 5Dear ReadersTwo things to tell you, as we graduallycome out of hibernation. The first thing to say,we are now back in printed form, so I do hope youare now returning back to a more structured way ofworking and reading this in the tea room! The second thing, something you will have alreadynoticed – the July edition of the Bulletin is muchsmaller than usual. There are a couple of reasons forthis, obviously while we have been in lockdown, wehave not been attending any conferences, meetings, seminars orworkshops in person, these have all been postponed or pushed intovirtual meetings. Most of which have been done at very short notice – socurrently there is nothing much to report. Additionally, because of theunscheduled changes to our lives, the Bulletin has begun to get verybehind in our print schedule, which we now need to get back as wereturn to printed format. Our August edition will also be small – but Iguarantee our September edition will be back to normal!Back with this edition, we do have some articles of interest to you – let’sstart with MySociety – the IAT membership database. Read pages 8-11 foran explanation of how this works and how you can use it. It is a charity with the purpose of advancing education and promoting excellence in the care and welfare of animals used in scienti c research.They have distributed over £180,000 so far in support of educational and welfare activities, with much of the money going towards educational activities which enhance individuals’ own knowledge and understanding. These courses range from short, specialist training to longer formal courses such as those leading to IAT quali cations.The criterion for obtaining money is that you are employed in care and welfare of animals used in science and the course or conference is related to your work. You must also show that you have sought a contribution for funding from another source – usually that would be an employer, but not always.From personal experience I know how diverse these bursaries can be, although many reports I have read come from people undertaking higher education on IAT courses, that is not the only application of the bursaries. I once applied for and was awarded a bursary, to help fund conference fees when I had to present work on lack of standardisation in Zebra sh welfare. In the end I never took the bursary, as the meeting was in the February and then in March, we were all in lockdown and the training spending I had planned for that year was never spent, so I could use it for that trip and the AS-ET money could be spent on somebody more deserving. I tell you this only because the convention was in Hawaii, so AS-ET can partially fund not just obvious training and education, but also that which is far more diverse. I would be a liar if I told you that whole trip was not only very educational and a chance to meet up with American colleagues but also as a bucket list holiday destination for me to travel to.Please look at the AS-ET webpages – https://www.as-et.org.uk/ and consider whether there is any training or educational event you might need some additional funding for. Bulletin6 • May 2023 West of Scotland Branch AGM and Seminar Held 24th February 2023Henry presenting...Joanne Main introducing the presentersAround twenty of us gathered for the presentations and AGM, it was great to see so many people attending. The meeting was quite informal and started with a few refreshments before the first presenter.The first speaker – Dr Henry McSorley, an immunologist from the University of Dundee gave a fascinating talk entitled ‘Worms to wheeze, how intestinal parasites suppress allergy’.
BulletinMay 2023 • 7 The T helper type II (TH2) immune inflammatory response is triggered by both allergens and parasitic infection and is sometimes referred to as the allergic response. These responses develop to kill parasitic worms; however, some parasitic worms can supress this immune response with interleukin 33 (IL33), a cytokine critical to this response. Historically, Helminth worm infection has been normal in human populations – witnessed in mummies from thousands of years ago and wild mammal populations. Sanitation has changed this in human populations and developed countries have suffered an epidemic of asthma, showing a negative correlation with having worms. This is explained in the Hygiene and Old Friends Hypotheses. Both of which state that early childhood exposure to certain organisms such as Helminth can protect again allergic reactions by developing the immune system and that lack of exposure in early childhood results in immune imbalances and subsequently disorders such as allergies and asthma. The model parasite – heligmosomoides polygyrus protects against pathology in mouse asthma models and supports these hypotheses.The questions are – how do worms protect against asthma and what molecules are involved? Finding out this information may help to create a drug to treat conditions connected to allegic reactions. To show this, worms cultured in vitro secreted HES (excretory / secretory products including lipids, carbs and extracellular vesicles). This was injected into one group of mice and a non-injected group were used as a control. Asthma was induced in both groups of mice using mould extract via an OVA protein (ovalbumin protein found in egg white) normally creates an immune response in two weeks. In the mice injected with HES, the pathology showed a much-reduced immune response.Further, IL33 was measured in epithelial cells of mice lungs, as it is released when these cells die – for example when killed by an allergen and then measurement of resistance to mechanical ventilation was read as measurement of lung function. Various proteins are produced by the worm which block IL33 receptors, therefore preventing and slowing cell death.Our second speaker Penny Hawkins from the RSPCA, known to many of us, gave an update on ‘Ending severe suffering – how techs can help’. 10% of UK laboratory animals are experiencing severe suffering and in context with the numbers of animals worldwide that number is huge. Since the start of actual severity recording there has been a 61% reduction in severe suffering due to the introduction of Earlier Humane End Points (EHEP), better screening, earlier disease stages, better communication, more involvement
Bulletin8 • May 2023 of technicians, the use of technology (for example temperature transponders) and the role and contribution of AWERBs.The causes of severe suffering have been identified as –1) The type of animal model used – for example some mouse models of diabetes. The RSPCA has written reports of models that are severe – which can be used as resources for those working with the models to identify refinements.2) Animal death as the end point – for example some fish in toxicology and other examples where a welfare assessment has been critical for preventing mortality in mammalian species.3) The cumulative severity of all procedures that an individual animal undergoes. The scruff test for lung tumour studies shows animals that take longer to recover after scruffing had a higher tumour burden which represents a better humane end point than weight loss. Further examples of cumulative suffering can include husbandry, handling, procedures, methods of killing and rehoming procedures.Penny encouraged everyone to start a conversation at their establishment about reviewing severe suffering and reminded us that the RSPCA is available if help is needed.Further information on this project can be viewed online at the Focus on severe suffering RSPCA website: https://focusonseveresuffering.co.uk/roadmap/Penny presenting...
BulletinMay 2023 • 9 Further, an NA3RsC compassion fatigue project has shown that severe suffering is linked to stress and burn out in Animal Technicians.Our final speaker was Fiachra McEnaney, a PhD student at University of Dundee who presented on his work – ‘The neglectful mother – how a model of early life stress induces depressed behaviour in adult mice’.Tianeptine is a clinical antidepressant and acts as an opioid agonist. In its effect it is not distinct from selective serotonin reuptake inhibitors (SSRIs) but it does not affect serotonin levels. Fiachra creates rodent models of depression based on adverse childhood experiences (ACE) – by providing limited bedding and nesting model in early life creating early life adversity (ELA) models.He recorded exits from the nest and time spent outside the nest – the ELA mother goes in and out of the nest and providing fragmented maternal care, resulting in a significant decrease in weight after weaning for ELA mice. Sociability is used as a measure of depression and so far, results have shown tianeptine improves sociability in ELA mice. He has also applied the 3Rs – using a less stressful test and observing multiple behaviours per mouse.Following these excellent presentations, we held the AGM, and then went onto the social part of the evening – where a great time was had by all!Reporter: Linda HoranWoS AGM

The aim was to understand the challenges and pitfalls all of us encounter every day during rodent surgeries. Your input will help to improve experimental surgery – thus reducing not only unnecessary suffering of animals, but also facilitating the generation of reproducible and reliable research data. The results of these surveys will hopefully be published soon.We are now working on a systematic review of available guidelines for goodsurgical practice during rodent surgery. We feel that internal guidelines / SOPs play a major role in the planning and conduction of rodent surgery. This Fellowship is valued at more than £3,000.
