kevinwhitaker.art7Advancing Parkinson’s and Lewy body dementia research at Sunnybrook While there is no cure for Lewy body dementia, researchers like Sunnybrook’s Dr. Mario Masellis are working hard to discover ways to control symptoms and improve quality of life. Kevin is benefitting from these novel approaches, which keep his symptoms at bay and allow him to express himself through art. “When I paint, I exist in a floating zone of creative thought. I am grateful to have this outlet,” he says. Kevin’s upcoming show, “Courage,” draws its title from the Tragically Hip song by the same name. “When I was diagnosed with Lewy body dementia six years ago, I learned that the average lifespan is five to eight years. Courage is a word I relate to every day.” It is with this sentiment that we thank you for engaging in Kevin’s fundraising campaign. You are making a difference in patients’ lives.Additional support would complete the funding required to establish a fellowship in Lewy body dementia research at the Hurvitz Brain Sciences Program, helping to accelerate the pace of discovery while training tomorrow’s leaders.The importance of research fellowsSiginificant and potentially practice-changing research into Lewy body dementia is already underway at Sunnybrook. While much of this work is led by renowned clinician-researchers like Dr. Masellis, research fellows are critical to moving the dial on discovery. Research fellowships offer academic health sciences centres like Sunnybrook the chance to attract and retain some of the best and brightest minds. ey are a prime example of Sunnybrook’s commitment, as an academic institution, to training the next generation of leaders. Here we have highlighted several groundbreaking research efforts by Dr. Masellis and his team to control symptoms, treat underlying conditions, gain precision and modify the course of Lewy body dementia. Discovering the blood pressure linkDr. Masellis has previously demonstrated a link between sudden drops in blood pressure caused by a sitting-to-standing movement (known as orthostatic hypotension) and drops in blood flow to the brain. e lower the blood pressure, the lower the blood flow – and crucially the more significant a patient’s cognitive impairment and visual difficulties. Building upon this discovery, Dr. Masellis and his team hypothesized that people who have Lewy body dementia with orthostatic hypotension will have worse cognition because the brain is not receiving optimal blood flow. e team performed arterial spin labelling, a very novel type of magnetic resonance imaging A diagnosis of Lewy body dementia may have forced Kevin Whitaker to retire early from his work as a Superior Court judge, but it didn’t change who he is. Smart, creative, passionate and thoughtful, Kevin is now channelling his formidable energy into painting, living a healthy lifestyle and fundraising to accelerate research at Sunnybrook. Almost half of them had significant orthostatic hypotension. Using a type of MRI scan that measures the structure of the brain, the group found that the patients with orthostatic hypotension also have higher rates of brain shrinkage. is affects several parts of the brain, including the medial temporal region, an important memory centre.A loss of brain cells and weakened connections in the memory centres of the brain “could explain why patients with orthostatic hypotension experience more cognitive impairment than those without sudden drops in blood pressure,” explains Dr. Masellis. is groundbreaking study is under review for publication at the prestigious journal Neurology. Discovering new treatmentse next step in this research is answering the question of whether various treatments to regulate blood pressure could impact cognition and cognitive decline. Another exciting area of research for Dr. Masellis and his team is precision medicine, which uses genetic analysis to determine how patients will repond to particular medications. In addition to his research, Dr. Masellis is a member of an international consortium charged with defining the criteria for the early signs and symptoms of Lewy body dementia that often indicate the onset of the disease before Lewy body dementia The second most common type of progressive dementia, which occurs when protein deposits, known as Lewy bodies, collect inside the brain’s nerve cells, interrupting messages. Current treatments temper symptoms, but there are no disease-modifying therapies yet available. You can change that by supporting vital research efforts. more diagnostically specific symptoms develop. Defining this warning stage is important, explains Dr. Masellis, “because by the time someone has full-blown Lewy body dementia, any drugs designed to target Lewy bodies, and thus modify the disease course, will be unlikely to work because the disease is too advanced.”e Holy Grail of research, says Dr. Masellis, is to actually learn how to modify the disease. To that end, clinical trials are starting that look at new therapies to target the alphasanuclein protein, the one responsible for Lewy body dementia.
