April 2024• 5BulletinKeep Reading!Carole As I write this, I have just returned from Congress, having had a great time in Aberdeen, one of the highlights, for me, being the karaoke! Of course, Congress – like all conferences also represents an excellent opportunity for CPD (continuous professional development), as well as the prospect of having great fun. Mark Gardiner (previous Honorary Member of the IAT) recently won an award certi cate from the Science Council, celebrating his outstanding professional development in science. He has written about winning and what CPD means to him on page 7. It is well worth a read, as it really explains the importance of this area of life to us all. Carrying on with the theme of CPD, many things can be considered – we have annual general meeting reports from both West of Scotland (WoS) and Surrey, Hampshire and Sussex (SH&S) Branches. Again, these would both be considered development opportunities – down in Guildford they had a presentation covering the history of Surrey Diagnostics, you can read this on pages 30-31. The WoS Branch had three presentations, all covering scienti c themes on pages 23 – 28. Indeed, when I read the precis of the talk on the effect of a novel δGABAA, I followed up with some more CPD of my own, as I read another paper to make sure I understood the subject.The Scotland Tech Day also had more presentations and therefore more opportunities for development but also included a talk on CLAST and training which is another source of continued education. You can see this report on pages 12 – 17.If you nd it dif cult to attend college, whatever the reason, or if you have already completed formal education, there are plenty of other sources to carryon learning – for example there are many papers you can access online, including Animal Technology and Welfare (ATW) at https://www.atwjournal.com/and more information on page 22. BulletinApril 2024 • 7Science Council Award for Outstanding Professional Development in ScienceIt was a real surprise receiving an award certifi cate from the Science Council for celebrating outstanding professional development in science.Especially, as continued professional development (CPD) to me, has always been about looking at the skills, knowledge and behaviours I require to enhance my job role and meet the challenges ahead. This could be reading papers, attending meetings, attending courses, professional discussions, listening to podcasts, etc. Anything really that fi lls in the gaps. For example, Sustainability and Carbon Net Zero is a big topic at the moment so I recently attended an Institute of Environmental Management and Assessment (IEMA) course and now have a better understanding of the language used and can have a positive input at meetings.The most important part of CPD, to me, after gathering new information is to refl ect on the content and ensure the gap has been fi lled, if not, then where to next? As I get older, time is precious and I fi nd it maddening when colleagues attend a course, only to place the material gained onto a shelf or nowadays into a fi le on a PC. No refl ection, no action plan and nothing gained. I also really feel that people get hung up on CPD and what to include. Actually, it is simply that you have learnt something new and then include it as part of your CPD. It really is that simple.Most of all, have fun collecting your CPD. As organisers we were delighted to see so many of you at papers, in workshops, in the trade hall and viewing the posters. It makes the hours of work pulling the event together really rewarding.Amongst other tasks as part of the Congress Committee, my main responsibility is “Posters”. Bulletin12 • April 2024 Scottish Tech Day 6th October 2023Setting up the Scottish Tech Day was a joint venture between three Scottish establishments: Dundee, Glasgow and Strathclyde Universities. The invitation went far and wide and you did not have to be Scottish to attend! Obviously, this was a great idea as we had around 60-70 technicians turn up for this in-person event. On arrival there was a delicious lunch laid on and a selection of trade stands to visit to see all the latest offerings from industry.We had four really interesting speakers discuss their work and a planned breakout enrichment discussion.Clicker Training in Mice Dr Nadine Baumgart, Expert for Laboratory Animal Sciences GV-SOLAS– PhD at the Institute for Immunology of the University Medical Center Mainz, Germany.– Since 2014 Head of a 3R research group at the (TARC (Translational Animal Research Centre) in the University Medical Center Mainz).– Currently Head of the TARCforce3R Center.– Animal Welfare Officer, BC Klinik Ludwigshafen.Refinement at RISE Research Institutes of Sweden – Stress-reduced handling of mice and rats Dr Matilda Bäckberg– Pharmacist and PhD in Neuroscience.– Director of the animal facility at RISE, which is a certified GLP toxicity laboratory.– RISE Research Institutes of Sweden conducts in vivo studies in the different steps of the preclinical drug development process (from exploratory studies to safety studies meeting regulatory criteria).

BulletinApril 2024 • 15Breakout groups at the Technicians Day Set-up to create discussion amongst different facilities. The topic of discussion largely focussed on the use of enrichment for the species at different establishments and consisted of the following questions:– What enrichment are you using and why?– Do you rotate this? Why and how often?– Introduction of novel objects?– Does your establishment have an enrichment strategy?– Ways of tackling single housing?– Importance of foraging?As mice were one of the largest groups used in animal research, this species was split into several groups and most groups agreed that similar enrichment was used at their establishments. The core enrichment required for each cage were tunnels/shelters such as dome homes, nesting, aspen chewsticks or aspen balls, with additional treats such as sunfl ower seeds, Nutella and the use of mezzanines, slings and Nombreros to enhance the use of space within the cage. These items however, were not rotated often or novel objects introduced. The groups also discussed that enrichment is provided as the standard and if there is any rotation of enrichment that this would likely be on a three-monthly basis.
Bulletin16 • April 2024 It was generally agreed across breakout groups that there was no enrichment strategy at their respective facilities, although there was one exception, some members observed that the lack of strategy was the sum of both custom and practice. Single housing, which can be detrimental to an animal’s health over time, was tackled by a number of different methods at different establishments. This included a requirement for scientifi c justifi cation of single housing being described on the PPL (legally, social animals must be housed in groups unless they are housed alone for welfare reasons); the use of companion males to maintain some social atmosphere where a male would otherwise be on his own; routine merging with females was also done, not only reduce single housing but to optimise the number of mice in the cage (a typical number of animals in a standard IVC cage, for example, being fi ve mice).Foraging is important in replicating natural behaviour exhibited in rodents and several groups agreed that it is necessary to replicate this in laboratory animals. This was achieved by the addition of items such as sunfl ower seeds, red peanuts, or meal worms to promote burrowing.Another species discussed during this session were rats, in which several facilities described their methods of enrichment. One mentioned the use of ‘double-decker’ cages which allow rats to climb and stand upright, another had a playpen room designed for rats (containing tunnels, lab coat sleeves, a dig box, pea fi shing and old racking boxes with ladders) into which rats go twice a week. Similarly, a different establishment had designed a playpen for rats to become more mobile and interact with new objects and mazes as a means of sensory enrichment and one facility had a veterinary student project which included the use of a water bath, bath towels and candy (but the rats were not so interested in the last of these). The general observation from this group was the preference for rats to climb.
BulletinApril 2024 • 17 Enrichment used for larger and lesser-known species being used at other facilities was mentioned and these included rabbits and guinea pigs which had the likes of pineapple or aspen chew blocks, large tunnels, rope pull cords, cardboard boxes, people and the radio but enrichment was generally not rotated. Other species discussed included cats (raised walkways and different levelled shelving, hanging beds and toys, sweeping brushes and people), Xenopus (plastic plants, lily pads and plastic hides) and bats (tea towels, hanging fruit, wicket baskets and buffalo worms).In conclusion, facilities have various means of providing enrichment to their animals with some implementing current ideas such as rat playpens/ playrooms to enhance enrichment further. However, it appeared that little evaluation had gone into whether what was provided was ‘the best’ for the animal. Going forward it is clear that a well-considered and suitably analysed enrichment strategy should be put into practice, to further develop this the NC3Rs, IAT and RSPCA https://nc3rs.org.uk/3rs-resources/evaluating-environmental-enrichment is a good resource to use. It is time we started to call many of the items currently put into the cages ‘species specific requirements’ (nesting material for mice and shelters for rats) as these are essential to meet the basic needs of the animal. We should then look to design an enrichment programme which adds to these. This month, the EDI Group is revisiting its “Let’s Talk” article on another protected characteristic, NeurodiversityLet’s talk about … Neurodiversity in the workplace“You probably can’t tell by looking at me, but my brain works very differently to yours. If you were to meet me, you’d probably notice that I’m tall and I have blonde hair. You might notice that I don’t often make eye contact. But you won’t see that I’m also extremely sensitive to bright lights and loud noises. I am hopeless at reading body language and I miss conversational cues. I love coding and enjoy solving problems. I struggle with changing environments, having to hot desk has a huge impact on me. I am not a neurotypical person, I am what is known as ‘neurodivergent’”What is Neurodiversity?Neurodiversity refers to the different ways the brain can work and interpret information. It highlights that people naturally think about things differently. We have different interests and motivations, and are naturally better at some things and poorer at others.Most people are neurotypical, meaning that the brain functions and processes information in the way society expects.However, it is estimated that around 1 in 7 people (more than 15% of people in the UK) are neurodivergent, meaning that the brain functions, learns and processes information differently. Neurodivergence includes Attention Deficit Disorders, Autism, Dyslexia and Dyspraxia. These bring strengths as well as difficulties.What does Neurodiversity mean?When we describe people as ‘neurodivergent’, then, we are talking about people who in one or several respects have a thinking style at the edges of one or more of these continuums, with – in the words of autistic author, speaker and educator Nick Walker – “A brain that functions in ways that diverge significantly from the dominant societal standards of “normal’. We need to understand that there is no standard brain.”Neurodiversity is, ultimately, a biological fact of the infinite variety of human neurocognition. Now, the same term ‘neurodiversity’ is also being used to represent a fast-growing sub-category of organisational diversity and inclusion that seeks to embrace and maximise the talents of people who think differently. If you were to meet me, you’d probably notice that I’m tall and I have blonde hair. You might notice that I don’t often make eye contact. But you won’t see that I’m also extremely sensitive to bright lights and loud noises. I am hopeless at reading body language and I miss conversational cues. I love coding and enjoy solving problems. I struggle with changing environments, having to hot desk has a huge impact on me. I am not a neurotypical person, I am what is known as ‘neurodivergent’”What is Neurodiversity?Neurodiversity refers to the different ways the brain can work and interpret information. It highlights that people naturally think about things differently. We have different interests and motivations, and are naturally better at some things and poorer at others.Most people are neurotypical, meaning that the brain functions and processes information in the way society expects.However, it is estimated that around 1 in 7 people (more than 15% of people in the UK) are neurodivergent, meaning that the brain functions, learns and processes information differently. Neurodivergence includes Attention Deficit Disorders, Autism, Dyslexia and Dyspraxia. These bring strengths as well as difficulties.What does Neurodiversity mean?When we describe people as ‘neurodivergent’, then, we are talking about people who in one or several respects have a thinking style at the edges of one or more of these continuums, with – in the words of autistic author, speaker and educator Nick Walker – “A brain that functions in ways that diverge significantly from the dominant societal standards of “normal’. We need to understand that there is no standard brain.”Neurodiversity is, ultimately, a biological fact of the infinite variety of human neurocognition. Now, the same term ‘neurodiversity’ is also being used to represent a fast-growing sub-category of organisational diversity and inclusion that seeks to embrace and maximise the talents of people who think differently. BulletinApril 2024 • 23 West of Scotland Branch Annual General Meeting (AGM) Friday 23rd February 2024 T his year, the West of Scotland Branch AGM was held at the University of Strathclyde and consisted of talks from researchers working at the University of Dundee and were hosted by our first presenter Joanne Mains who has been working as a senior technician at Dundee within the Medical School Resources Unit (MSRU). Thank you to IPS and Agenda for being our sponsors for this event.Joanne Mains: Procedural Specialist, University of DundeePresentation: Refinements of a Dual Catheter SurgeryJoanne’s research within the laboratory of Dr Li Kang is focussed on obesity and diabetes and discussed the refinements implemented in their dual catheter surgery being used for the glucose clamp technique. This is a gold standard method for assessing β-cell function, with different clamp techniques depending on the experimental conditions required. Previously, homemade bevelled catheters were used for ease during surgery but had the potential to cause post-surgical complications. Joanne was sent to Vanderbilt University to learn about new refinements which included pre-made sterile catheters with soft blunt ends, with one for the carotid arteries and jugular veins. These new catheters can reduce the risk of blood clots and strokes as well as a pre-made vascular access button with a felt base – the catheters themselves have small beads which help to judge the correct depth when catheterising. Additionally, a modified analgesic routine was used, which went from pre/post-operative subcutaneous injections of Rimadyl, to Rimadyl being given in water to improve animal recovery. Due to the movement of the animal during surgery (as the carotid artery and jugular vein are catheterised), full asepsis must be applied and Joanne explained their use of drapes and gowns, Opsite dressings and surgical gloves and of course a ‘dirty’ assistant to help with any non-sterile tasks that those performing surgery would avoid doing themselves. This is complex surgery and blood loss can be a concern. Joanne’s further studies will be starting soon and as part of these she will be learning how to use a cauteriser.
Bulletin24 • April 2024 Mattie Palowic: Principal Investigator, University of DundeePresentation: Refi nements of Mouse Models of CryptosporidiumFollowing Joanne, we had Principal Investigator Dr Mattie Palowic who explained her laboratory’s work on studying Cryptosporidium and how her team have implemented each of the 3R’s into their mouse models. Cryptosporidium is a deadly diarrhoeal disease, it is the number two killer in developing countries, ahead of HIV, Malaria and TB. Cryptosporidium cannot be cultured in vitro, as current in vitro techniques cannot replicate the complex environment of the gut so animal models have to be used to culture these parasites. Mattie explained the two main models used for Cryptosporidiumand how her laboratory has reduced the number of animals used in her studies (as only 50% were getting infected with GFP-Crypto, which help visualise the location of the parasites in the gut). After plotting the parasite yield, the model has been refi ned by culling the animals at the peak of their infection to maximise parasite collection and minimising the infection time in the acute models. They are currently working with a potential replacement by investigating organoid culture to replicate the intestines as much as possible.
BulletinApril 2024 • 25Yunkai Li: Postdoctoral Research Assistant, University of DundeePresentation: Investigating the effect of a novel δGABAA R enhancer on mouse behaviour and brain activityYunkai’s group presentation discussed the importance of the δGABAA receptor for maternal behaviour and its relationship to depressive behaviour, it has been linked in humans to post-natal depression. Mice with this receptor knockout showed a reduced pup survival rate compared to WT mice and reported data shows impaired forced-swim test and sucrose preference (tests for depression-like behaviours and anhedonia, respectively). The Positive Allosteric Modulator (PAM) known as DS-2 cannot cross the blood brain barrier, where Yunkai’s study is looking at another PAM referred to as “Drug X” and comparing it to the effects of GABAA agonist THIP; to determine whether “Drug X” exhibited antidepressant effects, the drug’s effects on brain activities and if it did it manage to cross the blood brain barrier and remain centrally active after administration. Both drugs produced behaviours lacking in balance and coordination while “Drug X” was shown to reduce locomotion in a dose-dependent manner compared to vehicle, whilst THIP produced a paradoxical response with an initial increase at 10mg/kg but producing a decrease at 30mg/kg. “Drug X” also promoted anti-anxiety effects during the elevated plus maze – used to test anxiety. The results, so far are promising for “Drug X” and Yunkai’s further studies will look at the effect of “Drug X” on emotional disorders by using an early-life adversity (ELA) model and using alternative methods for testing depression-like behaviours and anhedonia.Following the presentations there was a brief break before the AGM began with drinks and as part of Tech Month, there was a fun competition for who could make the best origami creation from the options of a dog, rabbit, or fi sh – with congratulations to Nicola Munro (AKA Lee Wheeler) for getting best prize.

Bulletin30 • April 2024 Surrey, Hampshire and Sussex (SH&S) Branch Annual General Meeting (AGM) Report 7th February 2024 T he SH&S Branch AGM took place at Surrey Diagnostics premises near Guildford. The meeting started with a presentation by General Manager Owen Hazelby. Owen explained that the original company was set up in 1996 by Professor John Bleby, a veterinary surgeon and former officer with the Royal Army Veterinary Corps (RAVC).Professor Bleby was Director of the MRC Laboratory Animals Centre (MRC LAC) at Carshalton. Under his leadership at the LAC, research and production activities prospered and the centre became an internationally recognised supplier of genetically and pathogenically standardised laboratory animals – most famously SPF cats, which Bleby himself developed. As a Vice President of the IAT and spokesman for the Research Defence Society (now Understanding Animal Research), Professor Bleby was a vocal supporter of the use of animals in medical research and he also campaigned to improve Animal Welfare in laboratories and on farms. Since it was established (at that time it only dealt with rodents), Surrey Diagnostics has expanded its activities into other species and disciplines. It is now an independent veterinary diagnostic laboratory offering personalised screening solutions and providing expert advice to clients across the UK and Europe. Owen detailed the company’s comprehensive array of services for scientific institutions and veterinary surgeries, encompassing serology, bacteriology, mycology, parasitology, molecular biology (PCR), environmental screening and allergen testing, across a diverse range of species. Surrey Diagnostics also provides environmental services to a broad spectrum of establishments including hospitals, schools and universities.He finished his presentation with an unusual test result, which was sperm in the anal canal of a group housed, male mouse!!
